Aston Villa defender Ezri Konsa and Nottingham Forest midfielder Morgan Gibbs-White have also pulled out of Lee Carsley’s party with the interim head coach deciding not to replace his withdrawals at this moment in time.
Captain Kane has been cleared to remain with the national team at St George’s Park after suffering ankle and groin issues in the weekend game against Eintracht Frankfurt.
The news will come as a key boost to Carsley, who is aiming to maintain his 100 percent record since replacing Gareth Southgate at the helm as he tries to land the boss’ job on a permanent basis.
However, the withdrawal of Manchester United star Mainoo will arrive as blow.

The central midfielder appeared to suffer a hamstring problem in the defeat to Tottenham on September 29 but did complete 85 minutes of Sunday’s goalless draw at Aston Villa.
It is understood Mainoo's withdrawal is precautionary.
Konsa also suffered a hamstring injury in the game at Villa Park, while Gibbs-White was forced off in their 1-1 draw at Chelsea with a suspected knee issue.
